Top 5 Partners
First of all, I have a tendency to partner-ship. Good and entertaining partnerships have a lot of the things I look for in romantic relationships: trust, banter, and there are usually guns involved.
1.) Charlie/Reese. What I love about this partnership is that it's an odd couple done really, really well. Reese doesn't like Charlie when they first met, and she doesn't like him when she first decides to keep him on the force. It's only later that they slowly start to become friends (and even Charlie, who was much more superficially friendly in the pilot, needed time to actually trust Dani). That slow building of the relationship made Reese's verbal smack-down of Charlie's old partner in the finale especially satisfying.
2.) Nathan/Audrey. These two are freaking adorable together. What I especially love is that they dynamic has a strong romantic and platonic vein. Sure, you have things like Nathan kissing the shapeshifter wearing Audrey's body and then shooting it (NOT LIKE I'VE WATCHED THAT A MILLION TIMES OR ANYTHING) and then you have Audrey getting Nathan dates and teasing him about his skill with woman and wearing the dumb clothes he buys her.
3.) Middleman/Wendy. They save the world together.
4.) Mary and Marshall. I don't ship these two romantically anymore (frankly, Marshall deserves someone who things he hangs the moon and isn't afraid to show him) but I love their gruff, abrasive friendship, and the way that they tease each other but have each other's back. The show makes it clear that not only are these two really effective when they work together but they have a friendship off company time.
5.) Touchstone and Sabriel. This is kind of pushing the definition of partners but they certainly weren't friends when Sabriel saved Touchstone's life and then they went and defeated a necromancer together.
Top 5 Vampires
1.) Caroline Forbes. God, what to say about Caroline? She's amazing and she tries so hard and she cares so much and gives 110% and just wants to be someone's first choice.
2.) Damon Salvatore. Look, I'm not proud of that, okay?
3.) William "Spike" The Bloody. a.k.a, the seminal snarky, semi-evil, fucking hot vampire of my fictional-tastes development.
4.) Jessica Hamby. The single, most-well constructed character arc on True Blood. There have been times when I've only watched the show to find out what was happening with her.

Mother Gothel. Her emotional abuse was almost too realistic for Disney, imho.
Mags Bennet. When even her own family isn't safe from her....
Darla. I think they did a fabulous job of showing hints of other aspects of Darla's personality while never back off on what a scary, fierce bitch she was. (Honorable Mention: Drusilla)
Evil, Alternate Universe Wendy Watson. The sign of a true hero is the fact that her alter-ego is out to destroy the world.

Okay, this is a slightly biased question, given that several shows have just wrapped, leaving me vaguely disappointed, and several shows have not yet started with new episodes, leaving me hopeful and optimistic.
So I'm going to answer this question in a completely un-5-things format.
Things I will be watching this fall: HIMYM, Nikita, TVD, Justified.
Things I am bouncing off the walls waiting for new episodes: Haven, True Blood, and Burn Notice.

I'd really love for them to play off of each other in-show, because despite/along with Raylan as a hurdle or whatever there's a lot of meat going on there - Winona's initial friendliness and curiousity and lack of jealousy ("did you screw things up with her?") Ava's discomfort in her home but near-raw honesty peeking through, the class tensions that I think were acknowledged a little bit in episode 12, the fledgling respect the text has implied lies between them...
I completely agree. I think there is a lot of things between Ava and Winona that could be stumbling blocks - the class tensions, Raylan – but likewise, there are a lot of things to explore and I think once they relaxed with each other a bit, they'd get along like a house on fire.
lso I want to write a fic where Winona wakes up Ava, getting her into bed and taking the handcuffs off (even though Raylan would have done it, shut up logic) and Ava is sleepy and possibly already a little hungover and Winona takes care of her.
THAT WOULD BE SO AWESOME.

I know there's a lot going on in the show like, at all times, and their storylines hardly intersect, but I really want them to get that chance. And I know the actresses do too - I loved that article you linked, where they talked about how awesome it was to play with/against the other women on the show.
